Quick tip: Sharpening images
Remember, you only sharpen the areas of the image where you want to be sharper. A sharp image doesn’t necessarily mean that all areas have to be sharp. If you make an image too sharp, you make it look like a printed page. Use this technique in the cropping section of this chapter where you see an image with some areas that are more blurry than others.

During ocular development, retinoic acid (RA) inhibits growth of precursors and immature retinal ganglion cells (RGCs), while inducing differentiation of other RGC types. The mechanisms of this dual effect on RGCs are not fully understood. In the present study, we investigated the role of the platelet-derived growth factor receptor (PDGFR) during RGC development in a developing vertebrate model, the zebrafish. Injections of RA into the developing zebrafish eye blocked the expression of the precocious zone marker of RGCs, orthopedia, in a manner dependent on the time of injection and the dose of RA. Using whole-mount in situ hybridization, we found that RA also inhibited the expression of the PDGF beta receptor (PDGFRbeta) in RGC precursors. Knockdown of PDGFRbeta expression mimicked the inhibition of orthopedia expression and the blockade of cell division by RA, implying that PDGFRbeta mediates the dual effect of RA on cell growth and division in zebrafish RGCs.Skeletal myoblast transplantation using intramuscular injection of collagen gel to mechanically enhance skeletal muscle regeneration in mice: a preliminary study.

The G protein family is the prototype of the seven-helix membrane-receptor family of proteins in the cell, and the main functional unit of the protein family is a heterotrimer of G protein subunit, the G protein alpha subunit (G alpha) and the G protein beta/gamma subunit (G beta/gamma). Several crystal structures of heterotrimeric G protein have been resolved and their intrinsic property has been investigated. To map the functional sites in G alpha, the crystal structures of guanine nucleotide bound with G alpha(o) complexed with the binary and ternary complex proteins (TCPs) have been solved. The ternary complex protein (TCP) is a cyclic nucleotide-activated and inhibitory protein. In the ternary complex, a cyclic nucleotide and the protein in the ternary complex are bound to G alpha(o). Thus, this structure has a nucleotide in the ligand binding pocket. This structure revealed the critical residues in the ligand binding pocket for GTP hydrolysis, and the effect of the ligand on the nucleotide binding pocket. Furthermore, the new crystallographic data with GDP bound to G alpha(o) revealed two GTP binding sites located at the residues distant to each other on G alpha(o). These crystal structures have been analyzed by sequence-based homology model in the G protein domain, and the functional sites including the nucleotide binding pocket, GTP sites and the P loops in G alpha(o) have been mapped. These findings will lead to a better understanding of the characteristic motif of the seven-helix family of proteins in the cell. was performed to optimize the extraction method.
